Pharmacology

This combination contains Paracetamol and Caffeine. Paracetamol has analgesic (pain-relieving) and antipyretic (fever-reducing) properties with mild anti-inflammatory effect. Caffeine is an alkaloid and a xanthine derivative similar to theophylline. It enhances the solubility and absorption of Paracetamol through intermolecular interaction, improving its effectiveness. In addition, Caffeine increases pain threshold and pain tolerance. It also increases cerebral vascular tone, which provides added benefit in the treatment of migraine and headache.

Dosage Administration

Adult dose: 1-2 tablets every 4-6 hours. Maximum dose: 8 tablets daily.

Child dose: Not recommended for children below 12 years.

Contraindications

Paracetamol is contraindicated in patients with severe renal impairment and hepatic diseases such as viral hepatitis. It is also contraindicated in patients with known hypersensitivity to paracetamol or caffeine.

Side Effects

Side effects of paracetamol are generally mild. However, hematological reactions such as thrombocytopenia, leukopenia, pancytopenia, neutropenia, and agranulocytosis have been reported. Occasionally, pancreatitis, skin rashes, and other allergic reactions may occur.

Pregnancy & Lactation

Pregnant women should consult a physician before taking Paracetamol & Caffeine. This combination may be used during breastfeeding.

Precautions & Warnings

Paracetamol & Caffeine should be used with caution in patients with hepatic or renal impairment and in those taking other hepatotoxic drugs. Prolonged use without medical supervision should be avoided.

Overdose Effects

Early symptoms of paracetamol overdose within 24 hours include pallor, nausea, vomiting, loss of appetite, and abdominal pain. Liver damage may appear 12 to 40 hours after ingestion. Disturbances in glucose metabolism and metabolic acidosis may also occur.
