Pancreatin

Indications

Pancreatin is used for the treatment of patients with:

  • Chronic pancreatitis
  • Cystic fibrosis (Mucoviscidosis)
  • Post partial or total pancreatectomy
  • Total gastrectomy
  • Steatorrhea
  • Somatostatinoma
  • Celiac disease
  • Other conditions associated with pancreatic insufficiency
Composition

Each enteric film-coated tablet contains 325 mg Pancreatin BP, providing not less than:

  • Amylase: 7800 units
  • Protease: 455 units
  • Lipase: 6500 units
Pharmacology

Pancreatin is an extract derived from mammalian pancreas containing digestive enzymes Amylase, Protease, and Lipase. It is formulated to disintegrate in the alkaline environment of the duodenum, releasing active enzymes. Amylase breaks down dietary starch into maltose, maltotriose, and α-dextrin. Protease cleaves peptide bonds in proteins and polypeptides, producing amino acids and oligopeptides. Lipase breaks down triglycerides into fatty acids and beta-monoglycerides. Pancreatin can convert at least 25 times its weight of starch into soluble carbohydrates and at least 25 times its weight of casein into proteoses within 5 minutes (equivalent to 150 times in 30 minutes).

Dosage Administration

2-3 tablets to be swallowed whole with little water immediately after or during meal. Or as directed by the physician

Interactions

Alkaline media may damage the enteric coating of Pancreatin. Therefore, to prevent premature release of the contents in the stomach, Pancreatin should not be taken together with antacids.

Contraindications

Pancreatin is contraindicated in patients with known hypersensitivity to the drug.

Side Effects

The most common side effects are gastrointestinal, including nausea, vomiting, and abdominal discomfort. Buccal and perianal soreness may occur, especially in infants. Hypersensitivity reactions such as sneezing, lacrimation, or skin rashes have also been reported.

Pregnancy & Lactation

Pancreatin should be used in pregnancy only if clearly needed. It is not known whether it is excreted in human breast milk. Since many drugs are excreted in milk, caution is advised when used during breastfeeding.

Precautions & Warnings

Patients with known allergy to animal protein should use this medicine with caution.

Overdose Effects

Excessive dose may cause a laxative effect. Systemic toxicity does not occur. Very high doses may lead to hyperuricemia and hyperuricosuria.
