Pemirolast acts by binding to histamine H1 receptors, thereby blocking the action of endogenous histamine and providing temporary relief from histamine-mediated allergic symptoms. It also inhibits antigen-induced calcium ion influx into mast cells by blocking calcium channels. In addition, Pemirolast reduces eosinophil chemotaxis into ocular tissues and prevents the release of inflammatory mediators from human eosinophils.
Dosage Administration
Adult: 1 to 2 drops in each affected eye four times daily.
Pediatric Use: Safety and effectiveness in pediatric patients below the age of 3 years have not been established.
Contraindications
Pemirolast ophthalmic solution is contraindicated in patients with known hypersensitivity to any of the ingredients of this product.
Side Effects
Burning sensation in the eye, dry eye, ocular discomfort, non-ocular allergy, sinusitis, headache, sneezing, or nasal congestion may occur.
Pregnancy & Lactation
Pregnancy: There are no adequate and well-controlled studies in pregnant women. Pemirolast ophthalmic solution should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus.
Lactation: It is not known whether Pemirolast potassium is excreted in human breast milk. Since many drugs are excreted in breast milk, caution should be exercised when administering it to nursing mothers.
Precautions & Warnings
To avoid contamination, the dropper tip should not touch the eyelids or surrounding areas. The bottle should be kept tightly closed when not in use. Patients should be advised not to wear contact lenses when the eye is red. Pemirolast should not be used for contact lens–related irritation. Patients should wait at least 10 minutes after using Pemirolast before inserting contact lenses.
