Chloramphenicol (Ophthalmic)
Generic MedicinePharmacology
Chloramphenicol is a broad-spectrum bacteriostatic antibiotic. It works by inhibiting bacterial protein synthesis, specifically by interfering with the transfer of activated amino acids from soluble RNA to the bacterial ribosome.
Dosage Administration
Adult and Children: Instill 1 or 2 drops in the conjunctival sac 4-6 times per day for the first 72 hours and then every 4 hours thereafter. Treatment should be continued for approximately 7 days, but should not be continued for more than 3 weeks without re-evaluation by the physician.
Interactions
Chloramphenicol may reduce or inhibit the activity of Chymotrypsin when both are administered at the same time.
Contraindications
Chloramphenicol should not be used in patients who have a known hypersensitivity or allergic reaction to chloramphenicol or to any component of this formulation.
Side Effects
With short-term topical use of Chloramphenicol, systemic adverse effects are generally uncommon. However, some patients may experience local reactions such as burning or stinging sensation, conjunctival redness (hyperemia), or allergic and inflammatory reactions. In rare cases, skin reactions like vesicular or maculopapular dermatitis and blood-related disorders (blood dyscrasias) have been reported.
Pregnancy & Lactation
The safety of Chloramphenicol during pregnancy and breastfeeding has not been fully established. Therefore, it should be used only when the physician considers it clearly necessary.
Precautions & Warnings
Chloramphenicol ophthalmic solution should not be used for minor eye infections or as a preventive treatment. Repeated courses and prolonged therapy should be avoided. Long-term use may increase the risk of blood dyscrasias, including granulocytopenia, thrombocytopenia, and moderate anemia.
Overdose Effects
Accidental swallowing of Chloramphenicol ophthalmic preparation is unlikely to cause significant toxicity because it contains only a small amount of the antibiotic.
Therapeutic Class
Ear Anti-infectives & Antiseptics, Eye Anti-infectives & Antiseptics, Antibiotics (Broad-spectrum)
