Meclizine has antiemetic, anticholinergic and antihistaminic properties. It decreases the sensitivity of the vestibular (labyrinthine) system. Its action may involve inhibition of pathways from the chemoreceptor trigger zone (CTZ) to the vomiting center (VC), as well as effects on peripheral nerves and other central nervous system centers. Meclizine has an onset of action within 30 to 60 minutes depending on the dose, and its duration of effect is 12 to 24 hours.
Dosage Administration
For the prevention and treatment of nausea, vomiting and vertigo associated with motion sickness, the usual oral dosage of meclizine hydrochloride in adults and children 12 years of age or older is 25-50 mg once daily.
Vertigo: for the control of vertigo associated with diseases affecting the vestibular system, the recommended dose is 25 to 100mg daily, in divided doses, depending upon clinical response.
Motion Sickness: The initial dose of 25 to 50mg should be taken one hour prior to embarkation for protection against motion sickness. Thereafter, the dose may be repeated every 24 hours for the duration of the journey.
Interactions
Additive CNS depression may occur when meclizine hydrochloride is used concomitantly with other CNS depressants, including barbiturates, tranquilizers, and alcohol.
Contraindications
Meclizine hydrochloride is contraindicated in individuals with a known history of hypersensitivity to the drug.
Side Effects
Drowsiness, dry mouth, and in rare cases blurred vision have been reported.
Pregnancy & Lactation
Pregnancy Category B by FDA. Safety for use in nursing mothers has not been established.
Precautions & Warnings
Due to its potential anticholinergic action, this drug should be used with caution in patients with asthma, glaucoma, or prostate enlargement.
