Indications

Antazoline + Tetryzoline is indicated for the temporary relief of symptoms associated with allergic conjunctivitis, including redness (conjunctival hyperemia), swelling (chemosis), and itching in adults and children aged 2 years and above.

Pharmacology

Antazoline is an antihistamine belonging to the ethylenediamine class and acts as a selective H1 receptor antagonist. It reduces allergic responses by blocking histamine-mediated effects such as capillary dilation, increased permeability, and smooth muscle contraction. Tetryzoline is a sympathomimetic agent with alpha-adrenergic activity that causes vasoconstriction, thereby reducing redness and swelling in the eye. While antihistamines are effective in relieving itching, they have limited effect on redness. Therefore, the combination of an antihistamine with a vasoconstrictor provides more comprehensive symptomatic relief in allergic eye conditions.

Dosage Administration

Adults and adolescents: 1 drop 2 to 3 times daily upto 14 days.

Children (Older than 2 years of age): No specific studies are available in this patient group. For possible systemic effects, the dosage should exceed to 1 to 2 drops per day.

Children (Below 2 years of age): Not to be used in children below 2 years of age.

The eye drop should not be used for longer periods than 14 days as this may cause rebound hyperemia and toxic follicular conjunctivitis

Interactions

Antazoline + Tetryzoline should not be used with monoamine oxidase inhibitors (MAOIs), as sympathomimetic agents may trigger a hypertensive crisis. Sedating antihistamines may increase the sedative effects of central nervous system depressants such as alcohol, hypnotics, opioid analgesics, anxiolytics, and antipsychotics. They may also enhance antimuscarinic effects when used with drugs like atropine and certain antidepressants.

Contraindications

This combination is contraindicated in patients with known hypersensitivity to Antazoline, Tetryzoline, or any component of the formulation. It is also contraindicated in patients receiving MAOI therapy.

Side Effects

The most frequently reported side effect is a mild and temporary burning or stinging sensation after application.

Pregnancy & Lactation

There are no adequate clinical data regarding use during pregnancy. This medication should be used only if the expected benefit outweighs potential risks. It is unknown whether the active ingredients are excreted in breast milk, so caution is advised during breastfeeding.

Precautions & Warnings

Use cautiously in elderly patients and in those with severe cardiovascular disorders such as arrhythmia, uncontrolled hypertension, or diabetes.

Overdose Effects

In case of topical overdose, the eye may be rinsed with clean warm water to remove excess medication.

Storage Conditions

Store at 15–25°C, protected from light. Use within 30 days after opening. Keep the bottle tightly closed and out of reach of children.
