Deferiprone is an iron-chelating agent that strongly binds to ferric ions (iron III). It forms stable, neutral complexes with iron in a 3:1 ratio (three deferiprone molecules to one iron ion), and these complexes remain stable across a wide range of pH levels. Deferiprone binds much more strongly to iron than to other metals like copper, aluminum, or zinc.
Dosage Administration
In adults and children over 6 years: this medicine may be given by mouth in doses of 25mg/kg three times daily. Doses above 100mg/kg daily are not recommended. Or, as directed by the registered physician.
Interactions
The safety of using Deferiprone together with vitamin C has not been formally evaluated. Since harmful interactions have been reported between deferoxamine and vitamin C, caution is advised when vitamin C is taken alongside Deferiprone. Deferiprone should not be used at the same time as antacids that contain aluminium.
Contraindications
Deferiprone should not be used in patients who are allergic or hypersensitive to the active ingredient or any of the other components (excipients) in the medicine.
Side Effects
Common side effects of deferiprone include gastrointestinal symptoms such as diarrhea, nausea, vomiting, and abdominal pain, which may sometimes require a temporary dose reduction. A reddish-brown discoloration of urine is also frequently observed. Other reported adverse effects include joint pain (arthralgia) and elevated liver enzyme levels.
Pregnancy & Lactation
Deferiprone is not recommended for use during pregnancy or breastfeeding.
Precautions & Warnings
Deferiprone has been associated with neutropenia, including severe cases such as agranulocytosis. Therefore, patients’ neutrophil counts should be checked every week. It should be used with caution in patients who have liver or kidney impairment.
